¾ Extraction of active ingredients from the tablet:

You will be assigned one of the four unknown analgesics

Take 1 tablet your assigned analgesic and grind between two sheets of weighing
paper.

Add to a 3 mL conical vial, add 2 mL methanol, cap and shake. Allow the solid to
settle; use a pipette to transfer the liquid to a centrifuge tube.

Repeat the methanol extraction two additional times and combine the methanol
extracts.

Centrifuge the mixture for 3 minutes (Make sure to balance the tubes using another
centrifuge tube of similar weight!)

Using a TLC spotter to transfer a small portion of your unknown analgesics in MeOH
onto blue litmus paper* and record your observation in Table 2. If there is no obvious
color change, take a new TLC spotter to transfer another small portion of your
unknown analgesics in MeOH onto a pink litmus paper* and record your observation
in the table below.

¾ Thin-layer Chromatography

Prepare your TLC chamber by adding 3 mL of ethyl acetate in it. Place a piece of
filter paper inside the chamber to saturate the air in the chamber with the solvent
vapors. Keep the cap closed.

Obtain two silica gel TLC plates from your instructor. Be careful not to bend the plate
because the flexible backing may cause some of the silica gel to flake off.
x
Take a pencil and draw a line with three equally spaced tick marks about 10 mm from
the bottom (This will be your baseline) and another line about 10 mm from the top
(This will be your solvent front line) for each TLC plate. These marks will eventually
become spotted with different samples, so label them accordingly: Use a pencil and
write U, Ace, Asp (representing Unknown, Aspirin, and Acetaminophen) above the
top pencil line on the first TLC plate, and write U, Ibu, and Nap ((representing
Unknown, Ibuprofen, and Naproxen Sodium) above the top pencil line on the second
TLC plate.

Using a TLC spotter, draw up a solution containing your unknown and spot it lightly
on both plates at the left tick marks. Allow the solvent to evaporate. Repeat for the
four known analgesic standards, in the right and middle tick marks of the two TLC
plates according to the writings above the top pencil lines of each plate.

Since all of these samples are UV active, check to make sure you have enough
analyte on the plate by looking at the plate under UV light. You should see a
black/blue spot on each tick mark.

Place one TLC plate into the chamber, and make sure that the solvent level is below
the baseline of your plate.
x
Develop the plate until the solvent front reaches the top pencil line. Take the plate
immediately out and allow the solvent to evaporate.

Place your plate under UV light. Use a pencil and draw a circle around each of the
spots you observe.
x
Repeat the process with the second TLC plate.
x
Calculate the retention factor (Rf) of each of the samples (Rf = danalyte/dsolvent front) and
complete the table below.
